华蟾素注射液基于阿片受体的镇痛机制研究

摘    要:目的: 探讨华蟾素注射液基于阿片受体的镇痛机制。 方法: 用辐射热和机械刺激使雌性昆明小鼠后足产生缩足反应,测定其缩足潜伏期(paw withdrawal latency,PWL)作为热辐射痛和机械性痛痛阈指标,观察外周阿片受体阻断剂Naloxone methiodide(纳洛酮的四价盐的衍生物,NAL-M)对华蟾素注射液升高小鼠PWL作用的影响。 结果: 外周阿片受体阻断剂NAL-M能够完全阻断华蟾素注射液升高小鼠PWL的作用,取消其镇痛作用。 结论: 华蟾素注射液的镇痛作用可能主要是通过外周的阿片受体介导的。

Abstract:Objective: To investigate the analgesic mechanism of cinobufagin injection based on opioid receptor. Method: Hind paw withdrawal latency of female Kunming mice was induced by radiant heat and mechanical stimuli.Moreover,the paw withdrawal latency (PWL) was measured to serve as indicator of pain threshold.Then,influence of naloxone methiodide,the peripheral opioid receptor antagonist,on PWL increase induced by cinobufagin injection in mice was investigated. Result: The analgesic effect and PWL increase induced by of cinobufagin injection could be completely blocked by naloxone methiodide. Conclusion: The peripheral opioid receptor may play a main role in the analgesic effect of cinobufagin injection.
