广防己、粉防己的肝肾毒性及代谢组学比较研究

摘    要:目的:比较广防己、粉防己血、尿代谢表型差异,结合常规毒理学指标探讨广防己、粉防己的毒性差异。方法:雄性SD大鼠70只随机抽取10只为空白对照组,其余60只分为正常对照组、广防己组、粉防己组,每组20只。广防己、粉防己组按8.1 g.kg-1.d-1灌胃给药,正常对照组灌服等体积蒸馏水。连续给药4周。取给药前、给药2周、给药4周3个时间点各组大鼠的血液和收集24 h尿液,分别测定血、尿1H-NMR谱,并做血生化检测;取肝、肾脏做组织病理学检查。结果:给药2周时,较之正常对照组,广防己、粉防己组BUN,广防己组AST均升高(P0.05);较之广防己组,粉防己组SCr升高(P0.05),广防己、粉防己组肾组织均出现明显病理改变;粉防己组肝组织出现病理改变。给药4周时,较之正常对照组,广防己、粉防己组BUN,ALT,AST,广防己组Scr均显著升高(P0.01),肝、肾组织病理改变加重;较之广防己组,粉防己组BUN,SCr,AST均升高(P0.05)。广防己、粉防己组大鼠尿液中主要变化代谢物有柠檬酸,2-酮戊二酸,牛磺酸,马尿酸盐,TMAO,肌酐;血液中主要变化代谢物有不饱和脂肪酸,3-羟基丁酸,丙酮,乙酰乙酸,肌酐,NAC,OAC。结论:广防己、粉防己均可引起肝、肾损害,且随给药时间延长损伤加重;粉防己的肝、肾毒性较广防己重。
