银杏叶提取物及银杏内酯B对体外培养大鼠神经元损伤的保护作用比较

摘    要:目的:经预处理给药模式观察了银杏叶提取物(extract ofGinkgo biloba,EGb761)及其单体银杏内酯B(ginkgol-ide B,GB)对神经元损伤的影响,以探讨EGb761及GB在抗兴奋毒性神经损害中的应用价值与实施途径。方法:原代培养新生Sprague-Dawley(SD)大鼠的海马神经元,建立谷氨酸诱导的兴奋毒性模型。采用台盼蓝染色、凋亡神经元测定及乳酸脱氢酶(LDH)测定的方法观察不同剂量EGb761,GB的神经保护作用,并与NMDA(N-甲基-D-天门冬氨酸盐)受体非竞争性拮抗剂MK801的神经保护作用相比较。结果:EGb761,GB在不同剂量下均能不同程度的提高细胞存活率,降低凋亡率,减少LDH泄露,且在一定范围内保护作用呈剂量依赖的方式。EGb761,GB分别在100 mg.L-1,100μmol.L-1的剂量下有最佳的保护效果,但均弱于MK-801(10μmol.L-1),GB最佳剂量下保护效果优于EGb761。结论EGb761,GB对谷氨酸兴奋毒性损害有保护作用。在预处理给药模式的最佳剂量下,GB的保护效果优于EGb761,将其用于高危人群的预防干预可能有更大价值。
