IgG启动子探针对中药注射剂进行临床前安全性评价的研究

摘    要:目的:利用分子生物技术构建免疫球蛋白G(IgG)启动子调控绿色荧光蛋白(green fluorescent protein,GFP)转基因细胞,尝试以IgG基因表达起始为标准在临床前对中药注射剂进行其诱导II型变态反应的可能性预测。方法:构建IgG启动子调控绿色荧光蛋白表达的转基因细胞系,用葛根素,KCl,聚氧乙烯脱水山梨醇单油酸酯(吐温80)对该系统进行测试,并尝试用该系统对不同批次鱼腥草注射液进行筛选评价,观察统计各给药组中发光细胞数量的变化。结果:非离子表面活性剂吐温80对IgG表达启动过程有抑制作用,葛根素能显著激活IgG基因的表达,KCl对IgG表达无作用。所测批次鱼腥草注射液对IgG表达没有激活作用。结论:IgG启动子调控绿色荧光蛋白表达细胞模型能在IgG表达层次评价药物致敏性,吐温80能在转录水平上抑制IgG的表达,所测批次鱼腥草注射液在IgG转录层次上无诱导II型变态反应活性。
