粗叶悬钩子总生物碱对大鼠非酒精性脂肪肝病的抗氧化作用研究

摘    要:目的:研究粗叶悬钩子总生物碱对大鼠非酒精性脂肪肝(NAFLD)的防治作用,并初步探讨其作用机制.方法:60只SD大鼠,采用高脂饲料建立大鼠NAFLD模型,随机分为6组:正常对照组、模型组、东宝甘泰(CMCB 0.35 g·kg-1)组、粗叶悬钩子总生物碱(RAP)低、中、高剂量(0.36,0.72,1.44g·kg-1)组.给药4周后,计算大鼠肝脏指数,观察肝组织病理形态学变化,检测血清中AST,ALT,TNF-α,IL-6,MDA,SOD的量,肝脏组织中MDA,SOD,TNF-α,IL-6的表达变化.结果:RAP给药4周后,与模型组比较,大鼠肝组织脂肪变性明显减轻,高剂量组肝脏系数明显降低(P<0.05),中、高剂量组血清中AST,ALT,TNF-α,IL-6水平明显降低(P<0.05或P<0.01),高剂量组肝组织中SOD,MDA含量明显降低(P<0.05),3个剂量组肝组织中TNF-α,IL-6 mRNA的表达均降低(P<0.05或P<0.01).结论:RAP对大鼠NAFLD具有较好的防治作用,其 作用机制可能与清除氧自由基,减少脂质过氧化反应产物,抑制炎性细胞因子释放,减少炎症反应有关.
