摘    要:目的 观察艾塞那肽(exendin-4,EX4)对4-羟基壬烯醛(4-hydroxynonenal,4-HNE)诱导人视网膜色素上皮(retinal pigment epithelium,RPE)细胞氧化应激损伤的抑制作用。方法 体外培养人RPE细胞(ARPE-19),采用CCK-8法检测不同浓度EX-4处理后细胞增殖情况;利用倒置显微镜观察对照组、EX4处理组、4-HNE组和4-HNE+EX4处理组细胞形态变化;利用荧光显微镜观察各组细胞的活性氧自由基(reactive oxygen species,ROS)生成;运用流式细胞仪检测各组RPE细胞内ROS含量变化;化学比色法检测细胞中超氧化物歧化酶(superoxide dismutase,SOD)和丙二醛(malondialdehyde,MDA)的含量变化。结果 100.0 nmol·L-1 EX4对RPE细胞具有较强的保护作用,在此浓度下对10 μmol·L-1 4-HNE引起的氧化应激损伤具有较强的抑制作用(P<0.05)。荧光显微镜观察发现,与对照组相比,4-HNE组亮绿色高荧光信号明显增强;与4-HNE组相比,4-HNE+EX4处理组荧光信号强度明显减弱,细胞内ROS含量明显减少。流式细胞仪检测也发现,与对照组相比,4-HNE组细胞内ROS含量升高,差异有统计学意义(P<0.01);与4-HNE组比较,4-HNE+EX4处理组细胞内ROS含量明显降低(P<0.05)。4-HNE组SOD含量明显低于对照组,差异有统计学意义(P<0.01);与4-HNE组比较,4-HNE+EX4处理组SOD含量明显升高,差异有统计学意义(P<0.05)。与对照组相比,4-HNE组MDA含量明显升高(P<0.01);4-HNE+EX4处理组MDA含量较4-HNE组明显下降,差异有统计学意义(P<0.05)。结论 EX4对4-HNE诱导人RPE细胞的氧化应激损伤具有保护作用,对老年性黄斑变性的治疗具有一定的潜在价值。

Abstract:Objective To study the inhibitory effect of exendin-4 (EX4) on oxidative stress injury induced by 4-hydroxynonenal (4-HNE) in human retinal pigment epithelium (RPE) cells.Methods Human RPE cell line ARPE-19 cells were cultured in vitro, and CCK-8 assay was used to detect the proliferation of RPE cells treated by different concentrations of EX4.Inverted microscopy was used to observe the morphological changes of RPE cells in the control group, EX4 treatment group, 4-HNE group and 4-HNE+EX4 treatment group, fluorescence microscopy was used to observe the changes of reactive oxygen species (ROS) production and distribution in RPE cells, cellular ROS content in RPE cells was detected using flow cytometry,and changes of superoxide dismutase (SOD) and malondialdehyde (MDA) content in RPE cells of each group was measured using chemical colorimetry.Results Strong protective effect of 100 nmol·L-1 EX4 on RPE cells was detected (P<0.01), and 100 nmol·L-1 EX4 had significant inhibitory effect on oxidative stress injury induced by 10 μmol·L-1 4-HNE (P<0.05). Fluorescence microscopy showed 2’,7’-dichlorodi-hydrofluorescein fluorescence intensity in 4-HNE group was markedly enhanced compared with control group; while compared with 4-HNE group, those were significantly weakened in the 4-HNE+EX4 treatment group, and the content of ROS in cells was significantly reduced. Flow cytometry results also found that the content of ROS production in 4-HNE group was significantly increased compared with the control group (P<0.01), and the content of ROS production in 4-HNE+EX4 treatment group was significantly lower than that in 4-HNE group (P<0.05). The content of SOD in 4-HNE group was significantly lower than that in control group (P< 0.01). Compared with 4-HNE group, the content of SOD in 4-HNE+EX4 treatment group statistically increased (P<0.05). The MDA content of 4-HNE group was significantly increased than that of control group (P<0.01), and the MDA content of 4-HNE+EX4 group was significantly lower than that of 4-HNE group, and the difference was statistically significant (P< 0.05).Conclusion EX4 can inhibit oxidative stress injury induced by 4-HNE in human RPE cells, and have potential value for senile macular degeneration treatment.
