肺炎克雷伯菌的毒力特性及其对视网膜色素上皮细胞炎症相关因子表达的影响

摘    要:目的 比较高毒力肺炎克雷伯菌与经典肺炎克雷伯菌的毒力特性,探讨不同毒力的肺炎克雷伯菌对视网膜色素上皮细胞炎症相关因子表达的影响。方法 收集8株高毒力肺炎克雷伯菌及8株经典肺炎克雷伯菌,分为高毒力肺炎克雷伯菌组和经典肺炎克雷伯菌组,通过黏液丝实验、PCR扩增rmpA基因检测、细菌抗血清杀伤实验及细菌抗吞噬实验比较两组肺炎克雷伯菌的毒力特性;两组细菌分别感染视网膜色素上皮细胞,通过ELISA检测视网膜色素上皮细胞内单核细胞趋化蛋白-1(monocyte chemotactic protein-1,MCP-1)及白细胞介素-6(interleukin-6,IL-6)表达水平。结果 高毒力肺炎克雷伯菌组细菌的高黏液表型、rmpA基因表达水平、抗血清杀伤能力及抗巨噬细胞吞噬能力均显著高于经典肺炎克雷伯菌组;两组细菌分别感染视网膜色素上皮细胞后,高毒力肺炎克雷伯菌组MCP-1和IL-6含量分别为(1177.38±227.17)ng·L-1、(118.63±40.39)ng·L-1,经典肺炎克雷伯菌组分别为(669.50±62.01)ng·L-1、(53.75±14.04)ng·L-1;视网膜色素上皮细胞MCP-1和IL-6表达水平高毒力肺炎克雷伯菌组分别是经典肺炎克雷伯菌组的1.76倍和2.20倍,差异均有统计学意义(均为P<0.05)。结论 相对于经典肺炎克雷伯菌,高毒力肺炎克雷伯菌具有高黏液表型、抗血清杀伤能力及抗巨噬细胞吞噬能力强的特性,且感染视网膜色素上皮细胞后,可引起更严重的炎症反应。

Abstract:Objective To compare the virulent characteristic of hypervirulent klebsiella pneumonia(hvKP)with classic klebsiella pneumonia (cKP) and to investigate the effect of KP on the expression of inflammatory cytokines of RPE cells.Methods Totally 8 strains of hvKP and 8 strains of cKP were collected and divided into hvKP group and cKP group.The virulence characteristics of KP were evaluated and compared by string test,PCR amplification of regulator of mucoid phenotype gene A (rmpA) gene test,antiserum trial and survival within macrophages experiment.Then the secretion level of monocyte chemotactic protein-1(MCP-1) and interleukin-6(IL-6) of PER cells infected by KP strains of these two groups were detected separately by enzyme linked immunosorbent assay (ELISA).Results The hypermucoid phenotype,the level of rmpA gene expression,the ability of antiserum killing,and the survival rate within microphages in the hvKP group were statistically significant higher than those in the cKP group.After RPE cells were infected with KP,the levels of MCP-1 and IL-6 in the supernatant were (1177.38±227.17)ng·L-1and (118.63±40.39)ng·L-1 in the hvKP group,seperatively,and (669.50±62.01)ng·L-1 and (53.75±14.04)ng·L-1 in the cKP group respectively.The levels of MCP-1 and IL-6 secreted by RPE cells in the hvKP group were 1.76 and 2.20 times accordingly higher than those in the cKP group,and both approached statistically significant difference (both P<0.05).Conclusion hvKP has the characteristics of hypermucoid phenotype,stronger antiserum ability and high survival rate within macrophages phagocytosis when compared with the cKP,and can induce more serious inflammatory reaction as RPE cells are infected with KP.
