热休克蛋白72(HSP72)对大鼠青光眼模型视网膜神经节细胞和视神经的保护作用

摘    要:目的探讨热休克蛋白72(heat shock protein 72,HSP72)对大鼠青光眼模型视网膜神经节细胞(retinal ganglial cells,RGCs)和视神经的保护作用。方法选取Wistar大鼠46只,采用随机数字表法将所有大鼠分为正常对照组(6只)和实验组(40只),实验组40只大鼠中8只不作处理(实验对照组),在制作青光眼模型后2 d,给予16只大鼠热休克反应处理(热休克反应组),16只大鼠硫酸锌腹腔注射(硫酸锌注射组)。观察及比较治疗前后各组大鼠眼压、RGCs中HSP72抗体含量、RGCs平均密度。结果激光后3 d、7 d、14 d、28 d,各组大鼠右眼眼压均较激光前有所上升,且各组间差异均无统计学意义(均为P>0.05)。正常对照组大鼠不同时间点HSP72抗体在RGCs中无表达,激光后3 d实验组各组大鼠RGCs中HSP72抗体含量缓慢上升,7 d、14 d达高峰,此后逐渐下降至接近正常水平。激光后,实验组各组大鼠RGCs中HSP72抗体含量显著高于正常对照组,且热休克反应组RGCs中HSP72抗体含量均高于实验对照组,低于硫酸锌注射组,差异均有统计学意义(均为P<0.05)。激光后3 d、7 d、14 d、28 d实验组各组大鼠RGCs平均密度均明显低于正常对照组,且热休克反应组RGCs平均密度显著高于实验对照组和硫酸锌注射组,差异均有统计学意义(均为P<0.05)。结论热休克蛋白反应可在青光眼模型大鼠RGCs中诱导HSP72的生成,且对青光眼性RGCs和视神经具有保护作用。

Abstract:Objective To investigate the protective effect of heat shock protein 72 (HSP72)on retinal ganglion cells (RGCs)and optic nerve in rat model of glaucoma.Methods Totally 46 Wistar rats were randomly divided into control group (n=6) and experimental group (n=40).In the experimental group,sixteen rats were treated with heat shock reaction (heat shock response group) and 16 rats were intraperitoneally injected with zinc sulfate (zinc sulfate injection group) 2 days after the establishment of rat glaucoma model,and the rest 8 rats were untreated (experimental control group).Intraocular pressure (IOP),HSP72 antibody content and mean RGCs density were observed and compared before and after treatment.Results The IOP of right eye in each group was higher than that before laser on the 3rd day,7th day,14th day and 28th day after laser,and there was no significant difference between each group (P>0.05).There was no HSP72 antibody expression in RGCs at different time points in the normal control group.The HSP72 antibody content in RGCs in the experimental group increased slowly at 3 days after laser irradiation and reached its peak at 14 days after laser irradiation,and then gradually decreased to the normal level.The content of HSP72 antibody in RGCs of experimental group after laser treatment was significantly higher than that in normal control group,and the HSP72 antibody content in RGCs of heat shock reaction group was significantly higher than that in experimental control group and zinc sulfate injection group (both P<0.05).The average density of RGCs in the experimental group was significantly lower than that in the normal group on the 14th and 28th days after laser irradiation,and the mean density of RGCs in the heat shock response group was significantly higher than that in the experimental control group and the zinc sulfate injection group (all P<0.05).Conclusion Heat shock protein reaction can induce HSP72 production in RGCs of glaucoma model rats and has protective effect on glaucoma RGCs and optic nerve injury.
