2016—2019年北京市海淀区副溶血性弧菌血清型、毒力基因及分子分型研究

摘    要:目的 综合分析北京市海淀区副溶血性弧菌血清型分布、毒力基因携带情况和PFGE分型结果,并找出其相关性,从而掌握海淀区副溶血性弧菌的流行特征。方法 对2016—2019年北京市海淀区检出的63株副溶血性弧菌进行血清分型、毒力基因检测和PFGE分型试验。结果 海淀区副溶血性弧菌最优势流行株的血清型为O3:K6,毒力基因组合为tlh+tdh+,占比69.8%(44/63);63株副溶血性弧菌经PFGE聚类分析得到37种带型,各带型之间相似度为84.8%~100.0%,其中,61株副溶血性弧菌聚集形成5个簇(带型相似度≥90%);4年间共出现了8次一定时间内(发病日期间隔≤90 d)分离得到的菌株血清型、毒力基因和PFGE带型均一致的情况。结论 海淀区副溶血性弧菌的污染来源相对固定且单一。副溶血性弧菌血清分型和PFGE之间无特殊相关性;毒力基因相同的菌株,PFGE带型相似度更高。

Serovar,virulence genes and molecular typing of Vibrio parahaemolyticus in Haidian District of Beijing from 2016 to 2019

Abstract:Abstract: Objective To analyze the sera distribution, virulence genes and PFGE typing of Vibrio parahaemolyticus?span>in Haidian District and discover the correlation between these factors in order to understand the epidemic characteristics of V. parahaemolyticus. Methods Serotyping, virulence gene detection and PFGE typing tests were performed on 63 strains of Vibrio parahaemolyticus detected in Haidian District, Beijing from 2016 to 2019. Results The most predominant epidemic strain of Vibrio parahaemolyticus in Haidian District was serotyped as O3:K6, and the virulence gene combination was tlh+tdh+, accounting for 69.8% (44/63). The 63 strains of Vibrio parahaemolyticus were analyzed by PFGE clustering to obtain 37band types, and the similarity between the band types was 84.8-100.0%, among which, 61 strains of Vibrio parahaemolyticus aggregated to form five clusters (band type similarity ≥90%). There were eight cases in which the serotypes, virulence genes and PFGE band types of the strains isolated within a certain period of time (interval between onset dates ≤90 days) were consistent during the 4 years. Conclusion The contamination source of V. parahaemolyticus in Haidian district is single and fixed. There is no special correlation between the serotype and PFGE pattern of V. parahaemolyticus. Strains with the same virulence genes have higher similarity of PFGE pattern.
