土槿皮乙酸对胰腺癌AsPC-1细胞增殖和凋亡的影响

摘    要:目的:观察土槿皮乙酸(pseudolaric acid B,PAB)对胰腺癌AsPC-1细胞增殖和凋亡的影响。方法:体外培养胰腺癌AsPC-1细胞,用不同浓度PAB作用不同时间后,MTT法检测PAB对AsPC-1细胞增殖的影响,流式细胞仪检测细胞凋亡率。结果:MTT结果表明PAB以时间和剂量依赖的方式抑制AsPC-1细胞生长,流式结果显示8.0μmol/L PAB处理AsPC-1 24小时后,凋亡率由对照组的8.19%升高至30.86%,差异有统计学意义(P<0.05),表明PAB可诱导胰腺癌细胞凋亡,呈剂量依赖性。结论:土槿皮乙酸以时间和剂量依赖的方式抑制胰腺癌AsPC-1细胞增殖并诱导其凋亡。

Abstract:Objective:To examine the effects of pseudolaric acid B (PAB) on the proliferation and apoptosis of human pancreatic carcinoma cell line ASPC-1.Methods:ASPC-1 cells cultured in vitro were treated with various concentrations (0.5,1.0,2.0,4.0,8.0μmol/L) of PAB at various intervals.Growth inhibition was analyzed by 3-(4,5-dimethylthiazol-2 yl)-2,5-diphenyltetrazolium bromide (MTT) assay.Apoptotic cells were detected by flow cytometry.Results:Pseudolaric acid B inhibited the proliferation of ASPC-1 cells in a time- and dose-dependent manner.The inhibition ratio was (4.53±3.32)% after the cells were treated with 0.5μmol/L PAB for 24h,and the ratio was (52.22±1.71)% after the cells were treated with 8.0μmol/L PAB for 72h.ASPC-1 cells treated with 8.0μmol/L PAB for 24h showed significantly enhanced apoptosis as compared with the control cells (8.19% vs 30.86%,P<0.05).Conclusion:PAB can dramatically suppress the ASPC-1 cell growth and induce cell apoptosis.
