摘    要:目的:探讨鼠双微体基因2(MDM2) T309G基因多态性与前列腺癌相关性。方法:用酚氯法提取全基因组DNA,应用聚合酶链反应、限制性内切酶的方法检测100例前列腺癌患者和100例健康男性的MDM2 T309G基因多态性。结果:病例组中G、T等位基因型频率分别为52.5%、47.5%,对照组中G、T等位基因型频率分别为40.5%、59.5%,MDM2 T309G基因型在两组的分布有显著性差异,TT基因型在对照组分布频率明显增高(P<0.05)。结论:MDM2 T309G基因型在病例组和对照组的分布存在明显差异,G等位基因与前列腺癌发病增加相关。

Abstract:Objective:To study the correlation of the single nucleotide polymorphism of MDM2 T309G and prostate cancer.Methods:The DNA was extracted by Phenol chloride.MDM2 T309G was examined by ploymerase chain reaction and restriction fragment length polymorphism in 100 cases of prostate cancer patients and 100 healthy people.Results:The frequency of G and T allele in the case group was 52.5% and 47.5%, in the control group was 40.5% and 59.5%. The distribution of MDM2 T309G genotype had significant differences in the two groups.The distribution of TT genotype in the case group was significantly lower in the control group(P<0.05).Conclusion:The distribution of MDM2 T309G genotype had significant differences in the two groups.G allele was associated with significantly increased risk for prostate cancer.
