小白菊内酯对甲状腺癌TPC-1细胞增殖、迁移和侵袭的影响及机制

摘    要:目的:研究小白菊内酯(parthenolide,PTL)对甲状腺癌TPC-1细胞增殖、迁移和侵袭的影响及其可能的机制。方法:应用0、2.5、5、10、20 μmol/L的PTL处理甲状腺癌TPC-1细胞24 h,CCK-8法检测不同浓度的PTL对TPC-1细胞增殖的影响;划痕实验和Transwell实验观察PTL对TPC-1细胞迁移和侵袭能力的影响;Western blotting检测E-cadherin、N-cadherin、Vimentin、MMP-9、TGF-β、Smad3、p-Smad3蛋白表达水平的变化。结果:CCK-8实验结果表明,PTL可抑制TPC-1细胞的增殖,并呈一定的浓度依赖性;划痕实验和Transwell实验表明,PTL可减弱TPC-1细胞的迁移和侵袭能力;Western blotting结果显示,PTL可上调E-cadherin的表达量,下调N-cadherin、Vimentin、MMP-9、TGF-β、p-Smad3的表达水平。结论:PTL以浓度依赖的方式抑制甲状腺癌TPC-1细胞增殖;PTL可能通过下调TGF-β的表达和抑制Smad3的磷酸化,逆转上皮间质转化(epithelial-mesenchymal transition,EMT)的发生,从而降低甲状腺癌TPC-1细胞的侵袭、迁移能力。

Abstract:Objective:To study the effect of parthenolide (PTL) on the proliferation,migration and invasion of TPC-1 cells in thyroid cancer and its possible mechanism.Methods:TPC-1 cells were treated with different concentrations of PTL (0,2.5,5,10,and 20 μmol/L) for 24 h,and the effect of on the proliferation was detected by the CCK-8 assay.We then examined the effect of PTL on cell migration and invasion via scratch and Transwell assays,respectively.The protein expression of E-cadherin,N-cadherin,Vimentin,MMP-9,TGF-β,Smad3 and p-Smad3 were detected by Western blotting.Results:CCK-8 assay results showed that the cell proliferation was suppressed in a concentration-dependent manner of PTL.Scratch and Transwell assays demonstrated that PTL significantly suppressed the migration and invasion ability of TPC-1 cells.Western blotting showed the E-cadherin expression was up-regulated,whereas N-cadherin,Vimentin,MMP-9,TGF-β,p-Smad3 expression were down-regulated following treatment with PTL.Conclusion:PTL inhibited cell proliferation of thyroid carcinoma TPC-1 cells in a concentration-dependent manner.PTL may reverse the occurrence of epithelial-mesenchymal transition by down-regulating the expression of TGF-β and inhibiting the phosphorylation of Smad3,reducing the invasion and migration ability of thyroid cancer TPC-1 cells.
