顺铂联合1,25(OH)2D3对肺癌细胞凋亡及VEGF、IL-6表达水平的影响

摘    要:目的:探讨顺铂联合1,25(OH)2D3对肺癌细胞凋亡及VEGF、IL-6表达水平的影响。方法:体外培养恶性胸水中肺癌细胞,经药物作用后CCK-8法测定细胞抑制率,流式细胞仪检测细胞周期和凋亡,应用化学发光仪检测细胞培养液中VEGF、IL-6表达水平。结果:不同浓度的1,25(OH)2D3、顺铂单独作用于肺癌细胞均有明显的抑制作用,两药联合表现为协同作用,与单药相比具有显著性差异(P<0.05)。流式细胞分析显示,经药物联合处理后的肺癌细胞的G0/G1期细胞数增多,诱导细胞凋亡(P<0.05)。顺铂、1,25(OH)2D3对VEGF的表达均有抑制作用,顺铂能够上调IL-6水平,1,25(OH)2D3对IL-6表达无显著作用。两药联合作用时能抑制VEGF、IL-6的表达,且较单药显著增强(P<0.05)。结论:顺铂联合1,25(OH)2D3能够抑制肺癌细胞生长,诱导细胞凋亡,降低了VEGF、IL-6的表达水平,可为肺癌提供新的治疗策略。

Abstract:Objective:To investigate the effect of cisplatin combined with 1,25(OH)2D3 on the apoptosis and the expression of VEGF and IL-6 in lung cancer cells.Methods:Lung cancer cells from malignant pleural effusion were cultured in vitro.The inhibition rate of tumor cells was measured by CCK-8 assay.The cell cycle and apoptosis were detected by flow cytometry.The expression level of VEGF and IL-6 in cell culture medium was detected by chemiluminescence.Results:Different concentrations of 1,25(OH)2D3 and cisplatin alone had obvious inhibitory effects on lung cancer cells,which increased with time and concentration increased (P<0.05).The combination of the two drugs showed synergistic effect,which was significantly different from that of single drug (P<0.05).Cell cycle analysis showed that the number of G0/G1 cells increased in the lung cancer cells treated by drug treatment and decreased in S and G2/M stages.DDP,1,25(OH)2D3 inhibited the expression of VEGF,DDP could decrease the level of IL-6,and 1,25(OH)2D3 had no significant effect on IL-6 expression.The combination of DDP and 1,25(OH)2D3 significantly induced the expression of VEGF and IL-6,and significantly enhanced (P<0.05) compared with the single drug.Conclusion:DDP combined with 1,25(OH)2D3 can inhibit the growth of lung cancer cells,induce apoptosis and reduce the expression level of VEGF and IL-6,can provide a new therapeutic strategy for lung cancer.
