摘    要:目的:探讨miR-21沉默对食管癌细胞增殖和凋亡的影响及其与β-catenin表达的关系。方法:通过脂质体转染法沉默食管癌Eca9706细胞株中的miR-21,采用荧光定量(RT-PCR)检测miR-21、β-catenin mRNA的相对表达水平;蛋白质印迹法(Western blot)检测β-catenin蛋白的相对表达水平。MTT法检测细胞增殖能力,流式细胞仪检测细胞凋亡和细胞周期。结果:转染48 h后,与对照组和NC组相比,miR-21组Eca9706细胞中miR-21、β-catenin mRNA和β-catenin蛋白的相对表达水平均明显降低。MTT检测miR-21组细胞的光密度值(OD值)较对照组和NC组明显下降。与NC组和对照组相比,流式细胞仪检测miR-21组中细胞的凋亡率明显上升,G0/G1期细胞所占比例明显升高,S期细胞所占比例明显下降。结论:沉默miR-21能够通过下调β-catenin的表达来抑制食管癌Eca9706细胞增殖及诱导其凋亡。

Abstract:Objective:To explore the influence of miR-21 silence on the proliferation and apoptosis of esophageal cancer cells and the relationship with β-catenin expression.Methods:miR-21 in Eca9706 esophagus carcinoma cell lines was silent by liposome transfection method,and use fluorescence quantitative(RT-PCR) and Western blot methods(Western blot) to detect the relative expression level of miR-21,β-catenin mRNA and β-catenin protein.The proliferation of esophageal cancer cells was detected by the tetramethazole blue method(MTT),and the apoptosis and cell cycle were detected by the flow cytometer.Results:The relative expression levels of miR-21,β-catenin mRNA and β-catenin protein in the miR-21 group of the Eca9706 cells were significantly lower than those of the control group and the NC group after 48 h transfecting.There was a significant decrease in the optical density(OD) of the miR-21 cells group compared to the control group and the NC group which detected by MTT.Flow cytometry test results showed that cell apoptosis rate in miR-21 group increased and the cell proportion of G0/G1 phase increased significantly,and the cell proportion of S stage decreased obviously compared with NC group and the control group.Conclusion:The silent miR-21 was able to inhibit the proliferation of Eca9706 cells and induce the apoptosis by reducing the expression of the β-catenin.
