摘    要:目的:探究脐血清源性外泌体对肝癌Hep3B细胞活力的影响。方法:提取健康足月胎儿脐血清源性外泌体,加入细胞培养基孵育24小时,MTT检测细胞活力变化,倒置显微镜观察细胞形态变化,流式细胞术检测细胞凋亡,Western blot检测Cleaved-Caspase3表达变化。结果:脐血清源性外泌体能显著抑制肝癌Hep3B细胞活力,导致细胞发生形态学改变、凋亡细胞比例增加及活化的Caspase3水平上升。结论:脐血清源性外泌体能诱导肝癌细胞发生凋亡,具有潜在的抑制肿瘤的生物学活性。

Abstract:Objective:To investigate the effects of exosomes derived from human umbilical serum on Hep3B cell viability.Methods:Exosomes were isolated from umbilical serum of healthy full-term fetus and were added into cell medium for 24 h.Cell viability was evaluated using MTT assay,cell morphology was observed with invert microscope,cell apoptosis was analyzed by flow cytometry and the level of Cleaved-Caspase3 was detected by Western blot.Results:Exosomes derived from human umbilical serum can suppress Hep3B cell viability significantly,induce cell morphological changes,increase the proportion of apoptotic cells,accompanied with increased level of cleaved-Caspase3.Conclusion:Exosomes isolated from umbilical serum may induce apoptosis of hepatic cancer,acting with the bioactivity of tumor suppressor.
