安徽省马鞍山市市售臭干子中沙门菌的污染状况及其耐药性与分子流行特征研究

摘    要:  目的  了解安徽省马鞍山市市售臭干子及其生产原料中沙门菌的污染状况、血清型分布、耐药情况及分子流行特征,探讨不同的血清型、分子型与耐药性之间的关系。  方法  2020年6 — 9月采集马鞍山市市售臭干子及其生产加工的辅料等,并对其中的沙门菌进行检测;用微量肉汤稀释法对菌株进行药物敏感性试验;使用脉冲场凝胶电泳和多位点序列分型方法(multi-locus sequence typing,MLST)对分离的菌株进行分子分型;使用BioNumerics 7.6软件对PFGE图谱、药敏结果及MLST分型结果进行聚类分析。及环境均有沙门菌检出;29株沙门菌可分为6种血清型和1种未分型沙门菌;菌株对四环素、氨苄青霉素和链霉素的耐药率较高,分别为48.3%、34.5% 和 31.0%;菌株可分为9个PFGE型别和6个ST型别;用血清学、PFGE和MLST三种方法对分离株进行分型,所得的辛普森多样性指数分别为0.711、0.762和0.753。  结论  马鞍山市售臭干子沙门菌的污染率较高且其生产加工过程中存在沙门菌污染的情况,不同地区的分离株之间有一定的同源关系。

Abstract:  Objective  To investigate the contamination status, serotype distribution, drug resistance and molecular epidemiological characteristics of Salmonella strains isolated from stinky tofu sold in markets in Ma'anshan, Anhui, and explore the relationship between different serotype, molecular type and drug resistance of Salmonella.   Methods  Stinky tofu and the auxiliary materials of them were collected from the markets in Ma'anshan from June to September 2020 for Salmonella detection. The drug susceptibility of the strains isolated was tested by micro broth dilution. Pulsed field gel electrophoresis (PFGE) and multi-locus sequence typing (MLST) were used for molecular typing of the strains. The map of PFGE, drug susceptibility and MLST results were analyzed by using BioNumerics 7.6.   Results  The detection rate of Salmonella strains in stinky tofu was 10.3% (25/242) in Ma'anshan, Salmonella was detected in both auxiliary materials and process environment. The strains isolated could be divided into 6 serotypes and one unspecific serotype. The drug resistance rates to tetracycline, ampicillin and streptomycin were 48.3%, 34.5% and 31.0%, respectively. The strains could be divided into 9 PFGE types and 6 STs. The Simpson diversity index of the isolates were 0.711, 0.762 and 0.753 by serologic test, PFGE and MLST, respectively.   Conclusion  The contamination rate of Salmonella strains in stinky tofu sold in markets was high in Ma'anshan, Salmonella contamination existed in the process of stinky tofu. There were some homology relationships among isolates from different areas.
