区域淋巴结切除对小鼠结直肠肿瘤CEA、VEGF-C表达的影响

摘    要:目的:探讨区域淋巴结切除对小鼠结直肠肿瘤CEA、VEGF-C表达的影响。方法:将60只健康BALB/c小鼠分为3组,虚拟切口+PBS注射15只(A组),虚拟切口+CT26.WT注射30只(B组),模型组(区域淋巴结切除)+CT26.WT注射15只(C组)。再将B组分为B1组15只(未行区域淋巴结切除)、B2组15只(肿瘤埋植7天后行区域淋巴结切除)。用ELISA法检测各组小鼠血清CEA、VEGF-C浓度,并对各组小鼠的瘤块称重及测算体积。结果:A组、B1组、B2组、C组小鼠血清CEA浓度分别为(602.6±52.2)pg/mL、(640.3±37.6)pg/mL、(676.5±56.6)pg/mL、(698.9±45.7)pg/mL,四组小鼠血清VEGF-C浓度分别为(153.5±13.0)pg/mL、(164.6±14.6)pg/mL、(177.1±15.4)pg/mL、(186.0±17.2)pg/mL。B组、C组的小鼠血清CEA、VEGF-C浓度相比A组均有不同程度的升高(P<0.05);与B1组相比,B2组、C组小鼠血清CEA、VEGF-C浓度均升高(P<0.05);瘤块重量及体积均明显增大(P<0.05)。结论:区域淋巴结切除可能会使小鼠肿瘤CEA及VEGF-C的表达上调,促进移植瘤的生长和侵袭。

Abstract:Objective:To investigate the effects of regional lymphadenectomy on expression of CEA and VEGF-C in mice colorectal tumors.Methods:60 healthy BALB/c mice were divided into three groups:Virtual incision+PBS injection,15 mice(group A),virtual incision+CT26.WT injection,30 mice(group B),and model group+CT26.WT injection,15 mice (group C).The model group mice were excised regional lymph nodes.The mice in group B were further divided into two groups:Group B1 (No regional lymphadenectomy was performed) and group B2(Regional lymphadenectomy was performed after 7 days of tumor implantation) with 15 mice in each group.The concentrations of CEA and VEGF-C in each group were detected by ELISA,and the tumor masses were weighed and measured.Results:The concentrations of CEA in group A,B1,B2 and C were (602.6±52.2) pg/mL,(640.3±37.6) pg/mL,(676.5±56.6) pg/mL and (698.9±45.7) pg/mL,VEGF-C concentrations in group A,B1,B2 and C were (153.5±13.0) pg/mL,(164.6±14.6) pg/mL,(177.1±15.4) pg/mL,(186.0±17.2) pg/mL.The concentrations of CEA and VEGF-C in group B and C were higher than those in group A(P<0.05).Compared with group B1,the concentrations of CEA and VEGF-C in group B2 and group C were increased(P<0.05).The weight and volume of tumor mass were larger than those in group B1(P<0.05).Conclusion:Regional lymphadenectomy can up-regulate the expression of CEA and VEGF-C in mouse tumors,promote the growth and invasion of transplanted tumor.
