TSHZ3在膀胱癌组织中的表达及其对膀胱癌细胞生物学功能的影响

摘    要:目的:采用生物信息学分析TSHZ3基因在膀胱癌组织中的表达水平,并通过体外实验探讨TSHZ3基因对膀胱癌细胞增殖、迁移和侵袭的影响,以及对上皮-间质转化相关蛋白的调控作用。方法:运用Oncomine和GEPIA数据库分析TSHZ3基因在膀胱癌组织中的表达水平,利用HPA数据库分析TSHZ3蛋白在正常膀胱和膀胱癌组织中的表达情况,并收集膀胱癌标本验证TSHZ3蛋白的表达。在膀胱癌T24细胞中转染TSHZ3质粒,CCK8法和克隆形成实验检测过表达TSHZ3基因对T24细胞增殖的影响,Transwell实验检测过表达TSHZ3基因对T24细胞迁移和侵袭的影响,Western blot检测过表达TSHZ3对上皮-间质转化相关蛋白的影响。结果:Oncomine和GEPIA数据库显示TSHZ3基因在膀胱癌组织中的表达水平降低。HPA数据库表明TSHZ3蛋白在正常膀胱尿路上皮组织中呈中等强度染色,而在膀胱尿路上皮癌组织中呈弱-中等强度染色。免疫组化结果显示TSHZ3在癌旁组织中呈现高表达,在膀胱癌组织中呈现低表达。过表达TSHZ3显著抑制T24细胞的增殖、迁移和侵袭能力。过表达TSHZ3增加E-cadherin蛋白的表达,降低N-cadherin和Vimentin蛋白的表达。结论:TSHZ3基因在膀胱癌中低表达,可作为抑癌基因抑制膀胱癌的发展。

Abstract:Objective:To analyze the expression level of TSHZ3 in bladder cancer tissues by bioinformatics analysis,and to explore the effect of TSHZ3 on the proliferation,migration and invasion of bladder cancer cells by in vitro experiments and the regulation role of TSHZ3 on epithelial-mesenchymal transition related proteins.Methods:Oncomine and GEPIA databases were used to analyze the expression level of TSHZ3 in bladder cancer tissues,and the human protein atlas (HPA) database was applied to analyze the protein expression of TSHZ3 in normal bladder tissues and bladder cancer tissues.The specimens of bladder cancer tissues were collected to verify the expression of TSHZ3 protein.TSHZ3 overexpression plasmid was transfected into bladder cancer T24 cells.Then,CCK8 assay and colony formation assay were performed to detect the effect of overexpression of TSHZ3 on the proliferation ability of T24 cells.Transwell assay was performed to investigate the impact of overexpression of TSHZ3 on the migration and invasion of T24 cells.Western blot was used to detect the effect of TSHZ3 overexpression on epithelial-mesenchymal transition related proteins.Results:Oncomine and GEPIA databases displayed that the expression level of TSHZ3 was decreased in bladder cancer tissues.HPA database showed that TSHZ3 protein was expressed in normal bladder urothelial tissues with moderate intensity staining,while it was expressed with weak to moderate intensity staining in bladder urothelial carcinoma tissues.Immunohistochemical results showed that TSHZ3 was highly expressed in paracancer tissues and low expressed in bladder cancer tissues.Overexpression of TSHZ3 obviously inhibited the proliferation,migration and invasion of T24 cells.And overexpression of TSHZ3 increased the expression of E-cadherin and decreased the expression of N-cadherin and Vimentin.Conclusion:TSHZ3 was downregulated in bladder cancer and might act as a tumor suppressor to inhibit the development of bladder cancer.
