摘    要:目的观察高浓度葡萄糖对人晶状体上皮细胞(SRA01/04)活性及沉默信号调节蛋白6(SIRT6)表达的影响。方法将SRA01/04细胞在不同葡萄糖浓度(5.5 mmol·L-1、15.5 mmol·L-1、25.5 mmol·L-1、35.5 mmol·L-1、45.5 mmol·L-1、55.5 mmol·L-1、75.5 mmol·L-1)培养基中培养48 h后,用光镜观察SRA01/04细胞形态;CCK-8试剂盒检测各组细胞的活性;Western blot检测各组细胞SIRT6蛋白的表达水平。然后将SRA01/04细胞用55.5mmol·L-1葡萄糖干预,培养不同时间(0 h、6 h、12 h、24 h、48 h、72 h)后,Western blot检测不同时间SRA01/04细胞SIRT6蛋白表达水平。免疫荧光技术检测对照组(5.5 mmol·L-1葡萄糖)和高糖组(55.5 mm...

Abstract:Objective To observe the effects of high concentrations of glucose on human-derived lens epithelial cells (SRA01/04) activity and silent information regulator 6 (SIRT6) expression.Methods SRA01/04 cells were cultured with different glucose concentrations (5.5 mmol·L-1, 15.5 mmol·L-1, 25.5 mmol·L-1, 35.5 mmol·L-1, 45.5 mmol·L-1, 55.5 mmol·L-1, 75.5 mmol·L-1) for 48 hours. The cell morphology was observed by light microscope, and the activity of cells in each group was detected by CCK-8 kit. The expression levels of SIRT6 protein in each group of cells were measured by Western blot. Then SRA01/04 cells were interfered with 55.5 mmol·L-1 glucose and cultured for different time (0 hour, 6 hours, 12 hours, 24 hours, 48 hours, 72 hours). The expression of SIRT6 protein in SRA01/04 cells at different time was detected by Western blot. Finally, immunofluorescence was used to detect the expression of SIRT6 protein in normal group (5.5 mmol·L-1 glucose) and high glucose group (55.5 mmol·L-1 glucose) cells.Results With the increase of glucose concentration, the number of SRA01/04 cells decreased gradually. The result of CCK-8 showed that the activity of SRA01/04 cells decreased gradually with the increase of glucose concentration compared with the control group, and the difference was statistically significant (P<0.05). The result of Western blot showed that the expression level of SIRT6 protein in SRA01/04 cells decreased in a dose-dependent and time-dependent manner with glucose, and the difference was statistically significant when comparison of 45.5 mmol·L-1, 55.5 mmol·L-1, 75.5 mmol·L-1 glucose group with 5.5 mmol·L-1 glucose group, and comparison of 24 hours group, 48 hours group and 72 hours group with 0 hour group (all P<0.05). The result of cellular immunofluorescence showed that the expression of SIRT6 in SRA01/04 cells was mainly in the nucleus, the expression of SIRT6 in SRA01/04 cells in high glucose group was significantly reduced, and the nuclei in high glucose group showed obvious nuclear pyknosis and fragmentation.Conclusion High concentration of glucose can reduce the proliferation activity of SRA01/04 cells and decrease the protein expression level of SIRT6 in SRA01/04 cells.
