叉头框蛋白M1在骨肉瘤组织中的表达及其与预后的相关性

摘    要:目的:探讨骨肉瘤组织中叉头框蛋白M1(forkhead box protein M1,FOXM1)的表达及其与预后的相关性。 方法:采用免疫组化SP法分别检测FOXM1在96例骨肉瘤组织和30例骨软骨瘤组织中的表达;分析FOXM1表达情况与骨肉瘤临床病理特征的关系;采用Log-Rank检验及Cox回归分析FOXM1与骨肉瘤患者预后的相关性,并通过Kaplan-Meier法绘制生存曲线。 结果:骨肉瘤组FOXM1高表达率高于对照组(P<0.05);骨肉瘤组织中FOXM1高表达与肿瘤临床分期和肺部转移相关(P<0.05);单因素分析显示,FOXM1蛋白高表达、肺部是否转移、临床分期是影响生存预后的独立因素(P<0.05);FOXM1蛋白高表达组的5年生存率为65.15%,低于低表达或无表达组的86.67%,差异有统计学意义(P<0.05);多因素分析显示,FOXM1蛋白高表达、临床分期和肺部转移是影响骨肉瘤患者生存预后的独立危险因素(P<0.05)。 结论:FOXM1蛋白在骨肉瘤组织中高表达,与患者的肺部转移及临床分期等病理特征有关,且高表达与较差预后相关。

Abstract:Objective:To explore the expression of forkhead box protein M1 (FOXM1) in osteosarcoma and its correlation with prognosis.Methods:Immunohistochemical SP method was used to detect the expression of FOXM1 in 96 cases of osteosarcoma and 30 cases of osteochondroma tissues.The relationship between FOXM1 expression and clinicopathological features of osteosarcoma was analyzed.Log-Rank test and Cox regression were used to analyze the correlation between FOXM1 and the prognosis of osteosarcoma patients,and survival curve was drawn by Kaplan-Meier method.Results:The high positive expression rate of FOXM1 in osteosarcoma group was higher than that in control group (P<0.05).The high positive expression of FOXM1 in osteosarcoma was associated with clinical stage and lung metastasis(P<0.05).Univariate analysis showed that high expression of FOXM1 protein,lung metastasis and clinical stage were independent factors affecting survival and prognosis (P<0.05).The 5-year survival rate of high expression group was 65.15%,which was lower than 86.67% in low or no expression group (P<0.05).Multivariate analysis showed that high expression of FOXM1 protein,clinical stage and lung metastasis were independent risk factors for survival and prognosis of osteosarcoma patients (P<0.05).Conclusion:FOXM1 protein is highly expressed in osteosarcoma tissues,which is associated with lung metastasis and clinical stage,and the high expression of FOXM1 protein is related to poor prognosis.
