原苏木素A通过ERK/c-Myc信号通路对人肺腺癌A549细胞放射敏感性的影响

摘    要:目的:探讨原苏木素A对人肺腺癌A549细胞放射敏感性的影响及其机制。方法:设置顺铂和空白对照,用MTT法测定不同浓度原苏木素A对人肺腺癌A549细胞的生长抑制情况;选取合适的药物浓度,采用集落形成实验,拟合细胞存活曲线,计算放射增敏比(SER);用Western-blot法对ERK、pERK、c-Myc、pc-Myc进行半定量分析。结果:原苏木素A对人肺腺癌A549细胞存在一定生长抑制作用,且存在剂量和时间依赖性,药物浓度越大、作用时间越长,其形态变化越明显。原苏木素A和外照射联合可增加其放射敏感性,且与浓度相关。原苏木素A联合外照射作用于A549细胞可通过下调ERK/c-Myc的表达增加其放射敏感性。结论:原苏木素A对人肺腺癌A549细胞具有生长抑制作用且存在药物剂量和作用时间依赖性;并可增加其放射敏感性;ERK/c-Myc信号通路为原苏木素A抑制A549细胞增殖的主要通路。

Abstract:Objective:To investigate the effects of progenistein A(PrA) on radiosensitivity of human lung adenocarcinoma A549 cells and its mechanism.Methods:Use MTT methods to measure the growth inhibition of human lung adenocarcinoma A549 cell by DDP and PrA of different concertrations.Select the appropriate drug concentration,adopt colony forming experiment,fit cell survival curve,to calculate SER.Semi-quantitative analysis of ERK,pERK,c-Myc,pc-Myc was performed by Western-blot method.Results:PrA had a certain growth inhibition effect on human lung adenocarcinoma A549 cells,which was dose-dependent and time-dependent.The larger the drug concentration and the longer the action time,the more obvious the morphological changed.The combination of PrA and external irradiation increased the radiosensitivity of A549 cells and was related to the concentration.PrA combined with external irradiation acting on A549 cells can increase its radiosensitivity by down-regulating the expression of ERK/c-Myc.Conclusion:PrA has the growth inhibition effect on A549 cells,which is dose-dependent and time-dependent.PrA can increase the radiosensitivity of A549 cells.ERK/c-Myc signaling pathway is the main pathway of PrA to inhibit the proliferation of A549 cells.
