CYP2C19基因多态性与云南汉族人群冠心病的相关性研究

摘    要:目的 冠心病(CHD)是一种致死率高,严重危害人类健康的疾病,我国该病发病率呈现上升趋势。遗传因素可增加冠心病风险。CYP2C19基因编码蛋白为细胞色素P450超家族成员之一,是人体重要的药物代谢酶。本研究旨在探讨CYP2C19基因多态性与中国云南汉族人群冠心病(CHD)的相关性。 方法 采用Sequenom MassARRAY®飞行质谱方法对云南省第二人民医院2012年9月-2015年9月根据知情同意原则收集的198例正常人(对照组)和514例冠心病患者(冠心病组)CYP2C19基因rs4986893、rs4244285、rs4917623三个单核苷多态(SNP)位点进行基因分型,Logistic回归法分析该基因多态性与冠心病的相关性。 结果 正常对照组和冠心病组3个位点的基因型和等位基因分布差异无统计学意义(P>0.05),Logistic回归分析校正年龄、性别、体重指数等,显示rs4986893位点携带A等位基因者患冠心病的风险增加显性模 AA/AG vs.GG:OR 1.07,95%CI (1.00~1.14),P=0.036]。 结论 CYP2C19基因多态性可增加云南汉族人群冠心病发生风险,但仍需大样本人群进一步研究。

Association of CYP2C19 gene polymorphism and coronary heart disease in Yunnan Han population

Abstract:Objective Coronary artery disease (CAD) represents a leading cause of morbidity and mortality worldwide,and genetic factors contribute to the development of this disease.The rate of coronary heart disease showed a rising trend in China.The CYP2C19 gene encodes a member of the cytochrome P450 superfamily of enzymes.It is an important drug metabolizing enzyme.We conducted a case-control study to assess the correlation between gene polymorphism of CYP2C19 and coronary heart disease (CHD) in Yunnan Han population. Methods One hundred and ninety-eight normal persons (control group) and 514 patients of coronary heart disease (CHD group) both written informed consents were recruited from September,2012 to September,2015.Three Polymorphisms rs4986893,rs4244285,rs4917623 in CYP2C19 gene were genotyped using the SEQUENOM Mass-ARRAY system.To investigate the correlation between gene polymorphism of cytochrome P450(CYP)2C19 and coronary heart disease using Logistic regression analysis. Results There were no significant differences in the genotypes and frequencies of alleles of CYP2C19 between the two groups (P>0.05).Logistic regression analysis showed that the rs4986893 A allele was associated with the increased risk of CHDthe dominant model:OR 1.07,95%CI(1.00-1.14),P=0.036]. Conclusion There is correlation between gene polymorphism of CYP2C19 and the risk of coronary heart disease in Yunnan Han Population.Future well designed large studies might be necessary.
