人脐带血间充质干细胞对人舌鳞癌移植瘤生长的影响及作用机制

摘    要:目的:探讨人脐带血间充质干细胞(HUC-MSCs)对人舌鳞癌细胞(Cal-27)移植瘤生长的影响及其相关特异性蛋白表达机制。方法:将Cal-27细胞移植到BALB/c裸小鼠皮下,待裸鼠肿瘤体积达到50 mm3成瘤标准后通过裸鼠尾静脉注射经GFP绿色荧光蛋白标记的HUC-MSCs,然后研究HUC-MSCs对预先建立的Cal-27细胞移植瘤生长状况的影响。为了阐明其发生机制,我们对具体阳性实验组及阳性对照组肿瘤组织的细胞增殖及凋亡相关蛋白Bcl-2、Bax、c-Myc、β-catenin的相对mRNA表达量进行了q-PCR分析实验。结果:在预先建立人舌鳞癌的体内模型中,通过尾静脉注射的经GFP绿色荧光蛋白标记的HUC-MSCs可以在肿瘤组织内富集并可抑制肿瘤生长。通过实时荧光定量PCR实验,我们发现阳性实验组相较于阳性对照组肿瘤组织细胞内Bax的mRNA表达量上调,Bcl-2、β-catenin、c-Myc的mRNA表达量下调。结论: HUC-MSCs通过促进Bax/Bcl-2形成同源二聚体以及抑制Wnt/β-catenin信号通路可抑制Cal-27细胞移植瘤生长。

Abstract:Objective:To investigate the effect of HUC-MSCs on human tongue squamous cell(Cal-27)carcinoma and the mechanism of its specific protein expression.Methods:Cal-27 cells were transplanted into BALB/c nude mice.After the tumor volume in nude mice was up to 50 mm3,we injected HUC-MSCs with GFP green fluorescent protein labeled by tail vein.Then we study the effect of HUC-MSCs on the growth of the transplanted tumor.In order to clarify the mechanism,we analyze the relative mRNA expression on cell proliferation and apoptosis proteins Bcl-2,Bax,c-Myc,β-catenin of the tumor tissue in the positive test group and the positive control group by q-PCR experimental.Results:In the model of human tongue squamous cell carcinoma in advance,HUC-MSCs labeled with GFP green fluorescent protein can be enriched in tumor tissues and inhibited tumor growth by intravenous injection of caudal vein.Through the q-PCR experiment,we found that compared with the positive control group,in the positive test group the mRNA expression of Bax was up-regulated,and the expression of Bcl-2,β-catenin and c-Myc was down regulated in tumor.Conclusion:HUC-MSCs inhibit tumor growth of Cal-27 transplanted tumor by promoting the formation of a homologous dimer of Bax/Bcl-2 and inhibiting the Wnt/β-catenin signaling pathway.
