摘    要:目的:探讨线粒体去乙酰化酶Sirt3基因对肝癌细胞增殖及侵袭的影响。方法:构建Sirt3载体,利用腺病毒感染方法建立Sirt3过表达的肝癌细胞(AdEasy-Sirt3),并设置相应对照(AdEasy-EGFP)。采用PCR与Western blotting实验分别鉴定病毒载体构建以及病毒感染后Sirt3蛋白过表达效果。采用EdU增殖实验与Transwell侵袭实验分别观察评价Sirt3对肝癌细胞的增殖与侵袭作用。结果:Sirt3腺病毒载体以及Sirt3蛋白过表达的肝癌细胞构建成功;与对照Ad-EGFP相比,Ad-Sirt3腺病毒感染的肝癌细胞增殖速度明显降低,差异具有统计学意义(P<0.05);Ad-EGFP组与Ad-Sirt3组肝癌细胞的侵袭能力差异亦具有统计学意义(P<0.01)。结论:过表达Sirt3可显著抑制肝癌细胞(HLF与HLE细胞株)增殖和侵袭,提示Sirt3可能是肝癌治疗的潜在靶点。

Abstract:Objective:To investigate the effect of Sirt3 on the proliferation and invasion of hepatocellular carcinoma cells.Methods:Sirt3 expression was increased using adenovirus,and Sirt3 overexpressed hepatocellular carcinoma cells were constructed.Expressions of Sirt3 in HCC cells were detected by Western blotting.The effect of Sirt3 on the proliferation of HCC cells was evaluated by EdU assay,and Transwell assay was used to evaluate cell invasion.Results:Sirt3 overexpressed hepatocellular carcinoma cells were successfully constructed.The cell growth rate in adenovirus-mediated Sirt3 overexpression group were markedly decreased compared to those in control group (P<0.05).The invasion potential of HCC cells differed significantly between Sirt3 overexpressed HCC cells and control cells (P<0.01).Conclusion:Sirt3 overexpression induced a significant decrease in HCC cell proliferation and invasion,which indicated that Sirt3 might be a potential therapeutic target for liver cancers.
