L1CAM在甲状腺乳头状癌组织及细胞中的表达及与临床病理特征的关系

摘    要:目的:检测甲状腺乳头状癌组织及细胞株中L1CAM表达,探讨其与临床病理特征的相关性及意义。方法:选取手术切除甲状腺乳头状癌组织、对应的癌旁组织及正常甲状腺组织标本各32例,甲状腺癌细胞株及正常甲状腺细胞株。采用Western blot及qRT-PCR检测组织及细胞株中L1CAM的表达,观察其与肿瘤部位、直径、数目、分化程度、淋巴结转移及AJCC分期等临床病理参数的关系。结果:L1CAM蛋白及mRNA在甲状腺乳头状癌组织中表达高于癌旁甲状腺组织,在正常甲状腺组织无表达(P<0.05);甲状腺乳头状癌组织中L1CAM蛋白水平与肿瘤直径、肿瘤数目、淋巴结转移、AJCC分期具有相关性(P均<0.05),肿瘤直径大于1 cm、肿瘤多发、淋巴结转移及AJCC分期Ⅲ-Ⅳ的甲状腺癌组织中L1CAM表达量高;L1CAM蛋白及mRNA在BHT101、B-CPAP甲状腺癌细胞株中均有表达,在正常甲状腺上皮细胞NTHY-ORI 3-1无表达,在BHT101中高于其他细胞株(P<0.05)。结论:L1CAM在甲状腺乳头状癌组织及细胞中表达上调,与甲状腺乳头状癌发展及转移恶性临床病理特征相关。

Abstract:Objective:To detect the expression of L1CAM in thyroid papillary cancer tissues and cell lines at protein and mRNA level,and discuss the association with clinicopathological characteristics.Methods:Thirty-two paired samples of thyroid papillary cancer tissues,para-cancer tissues,normal tissues,thyroid cancer cell lines and normal thyroid cell lines were selected and L1CAM expression was detected by Western blot and quantitative real-time polymerase chain reaction(qRT-PCR).The association between L1CAM protein levels and tumor location,diameter,number,differentiation,lymph node metastasis and AJCC stage was analyzed.Results:Expression of L1CAM protein and mRNA in thyroid papillary cancer tissues was higher than in adjacent normal thyroid tissues (P<0.05).L1CAM was not expressed in normal thyroid tissues.L1CAM protein level in thyroid tissues was correlated with tumor diameter,tumor number,lymph node metastasis,AJCC stage(P<0.05).L1CAM was high expression when tumor diameter over 1cm,multi-tumors,lymph node metastasis and Ⅲ-Ⅳ AJCC stage(P<0.05).L1CAM protein and mRNA was expressed in BHT101 and B-CPAP thyroid cancer cell-lines,and highest in BHT101 cells (P<0.05).L1CAM was not expressen in thyroid NTHY-ORI 3-1 cells.Conclusion:L1CAM is up-regulated in thyroid papillary tissue and cells,and associated with malignant clinical pathological characteristics of thyroid cancer development and metastasis.
