贵阳市禽类相关外环境1 013份标本中禽流感病毒检测结果分析

摘    要:目的 了解贵阳市禽类相关外环境H5、H7、H9亚型禽流感病毒的污染情况,为贵阳市人禽流感的防控提供依据。 方法 2015年10月- 2016年3月,每月定期采集贵阳市禽类相关外环境中不同类型的标本共1 013份,采用Real-time PCR方法对环境标本进行FluA、H5、H7、H9亚型禽流感病毒核酸检测,检测结果采用SPSS19.0软件进行分析。结果 贵阳市禽类相关外环境FluA阳性率为9.58%,H5、H9、H5+H9亚型禽流感阳性率分别为1.97%、4.94%、2.67%,未检测出H7亚型;不同监测点外环境FluA阳性率差异有统计学意义(χ2 = 66.741、P <0.05),活禽交易市场、数字化蛋禽交易中心FluA阳性率分别为8.99%、27.13%,禽类规模养殖场和禽类散养户集中地区未检出FluA;不同类型标本FluA阳性率差异无统计学意义(χ2 = 5.245,P >0.05),FluA在案板表面擦拭物、粪便、笼具表面擦拭物、洗禽污水、禽类饮水及其他标本中的阳性率分别为7.78%、8.82%、9.77%、7.64%、14.77%、14.29%;非中心城区活禽交易市场FluA阳性率高于中心城区(χ2 =50.534,P <0.05)。结论 贵阳市禽类相关外环境存在H5、H9亚型禽流感病毒污染,存在人感染高致病性禽流感的风险。

Abstract:Objective This work was to learn the distribution of avian influenza virus sub-type H5, H7 and H9 in the poultry related external environment in Guiyang and thus to provide references for the prevention and control of the human avian influenza of Guiyang. Methods A total of 1013 samples were monthly collected from the poultry related external environment in Guiyang from October 2015 to March 2016.While Real-time PCR was employed to detect the nucleic acid of avian influenza virus Flu A and H5,H7, H9 sub-types in environment samples. The data were analyzed by SPSS 19.0 software. Results The positive rate of FluA in poultry related external environment in Guiyang was 9.58%. H5, H9, H5 and H9 subtype of avian influenza were 1.97%, 1.97%, 4.94%, respectively, but H7 subtypes was not detected. Detection rate differed significantly with monitoring sites (χ2=66.741,P<0.05). The positive rate of samples from live poultry market and Digital egg trading center were 8.99%, and 27.13%, respectively. The external environment samples of large scale poultry farms and free-range poultry farmers concentrated areas were negative. There was significant difference in detection rates of these samples (χ2=5.245,P>0.05). The positive rate of FluA on board surface wipe, feces, basket surface cleaning, washing poultry water, drinking water and other specimens were 7.78%, 8.82%, 9.77%, 8.82%, 14.77% and 14.29%, respectively. The live poultry markets at suburban area had higher detection rate than the central area of the city (χ2=50.534,P<0.05). Conclusion Avian influenza virus H5 and H9 sub-types were found in poultry related external environment in Guiyang, presenting high risk of avian influenza to human.
