摘    要:目的 探讨瘦素对人成骨样细胞MG63 Ⅰ型胶原A1基因表达的影响.方法 MG63细胞以3个不同浓度的瘦素(10~(-8)~10~(-6) mol/L)分别干预24、48、72 h,用实时荧光定量PCR法检测MG63细胞Ⅰ型胶原A1基因 mRNA的表达量,分析量效关系和时效关系,以17β-雌二醇作为阳性对照.结果 瘦素可上调MG63细胞Ⅰ型胶原A1基因 mRNA的表达,并存在浓度依赖和时间依赖效应,其最佳浓度为10~(-7) mol/L,最佳作用时间点为72 h.17β-雌二醇则以10~(-7) mol/L浓度组在24 h表达为最强.结论 瘦素可上调MG63细胞Ⅰ型胶原A1基因mRNA的表达,其作用较17β-雌二醇持久和滞后.

Effects of leptin on expression of alpha1 (Ⅰ) collagen gene in human osteoblast-like cell line MG63 cells
Abstract:Objective To examine the effects of leptin on alpha1 (Ⅰ) collagen gene expression in human osteoblast-like cell line MG63. Methods MG63 cells were incubated with different doses of leptin (10~(-8)、10~(-7)、10~(-6) mol/L) for 24、48、72 h. The real time flourescence quantitive PCR (FQ-PCR) was performed to detect the mRNA expression of alpha1 (Ⅰ) collagen gene in MG63 cells, with 17β-E_2 as positive control. Results Alpha1 (Ⅰ) collagen gene expression regulated by leptin showed a dose-dependent and time-dependent manners, with the best effect concentration at 10~(-7) mol/L, and maximal expression at 72 h. As positive control, 17β-E_2 also reached its maximal effect in 10~(-7) mol/L group at 24 h. Conclusions Leptin up-regulated alpha1 (I) collagen gene expression in MG63, which is more enduring and later than that of 17β-E_2.
