清脑宣窍滴丸对实验性急性脑缺血损伤大鼠一氧化氮及其合酶的影响

摘    要:目的:研究清脑宣窍滴丸对急性脑缺血/灌注损伤引起的一氧化氮(NO)含量和一氧化氮合酶(NOS)活力的改变及机制。方法:采用线栓法致大鼠大脑中动脉缺血再灌注(MCAO)模型,测定大鼠手术侧脑匀浆中NO含量及结构型一氧化氮合酶(cNOS)、诱导型一氧化氮合酶(iNOS)、总NOS活力。结果:清脑宣窍滴丸45,90,180mg.kg-1可明显降低MCAO大鼠患侧脑匀浆中NO含量,180mg.kg-1可明显降低术侧大脑组织升高的总NOS,cNOS和iNOS活力(P<0.05);90mg.kg-1也可明显降低升高的总NOS活力(P<0.01)。45mg.kg-1明显降低升高的总NOS活力(P<0.05)。结论:清脑宣窍滴丸可能通过减少NO含量,降低iNOS的活性来防治脑缺血/再灌注损伤。

Abstract:Objective: To study the role and mechanism of Qing-nao-xuanqiao Dropping pill(QNXQD) on acute cerebral ischemic injury.Methods:The middle cerebral artery occlusion(MCAO)model induced by inserting a monofilament suture was adopted.The content of NO,and the activity of cNOS,iNOS,total NOS in brain homogenate was determined.Results: QNXQD 45,90,180 mg·kg-1,can significantly reduce the content of NO in the rat brain homogenate of the troubled side;180 mg·kg-1 can significantly decrease the increased cNOS,iNOS;total NOS activity in the side of the brain surgery(P<0.05);90 mg·kg-1 can significantly decrease the increased total NOS activity(P<0.01);and 45 mg·kg-1 can significantly decrease the increased total NOS activity(P<0.01).Conclusion: QNXQD has protective effect on cerebral ischemia and the possible mechanism may be related to reducing content of NO,decreasing the activity of cNOS,iNOS,total NOS.
