沉默ZIC1基因联合氧化苦参碱对子宫内膜癌KLE细胞生物学行为的影响

摘    要:目的:探讨沉默ZIC1基因转染联合氧化苦参碱对人子宫内膜癌KLE细胞生物学行为的影响。方法:将慢病毒载体pLV-shZIC1-PGK-Puro稳定转染人子宫内膜癌KLE细胞,通过Western blot法检测转染后ZIC1蛋白的表达水平。通过MTT法筛选出氧化苦参碱的IC50浓度后,分为空载不加氧化苦参碱组(A组)、空载加氧化苦参碱(B组)、转染不加氧化苦参碱组(C组)及转染加氧化苦参碱组(D组),分别采用MTT法检测对KLE细胞黏附的影响,划痕实验检测细胞的迁移能力,流式细胞术检测各组细胞的凋亡率。结果:子宫内膜癌细胞转染shZIC1后,ZIC1蛋白表达明显下调。氧化苦参碱或沉默ZIC1单独作用均可抑制KLE人子宫内膜癌细胞的黏附、迁移及促进细胞凋亡(P<0.05),且在抑制细胞迁移上存在时间依赖性。沉默ZIC1联合氧化苦参碱可影响子宫内膜癌KLE细胞的凋亡和侵袭,差异有统计学意义(P<0.05)。结论:沉默ZIC1与氧化苦参碱对子宫内膜癌的抑制存在协同作用,联合应用可明显增强对人子宫内膜癌KLE细胞的抑癌效果。

Abstract:Objective:To investigate the influence of ZIC1 gene combined with oxymatrine on biological behavior of KLE cells in human endometrial cancer.Methods:Lentivirus vector pLV-shZIC1-PGK-Puro was transfected stably into KLE cell,Western blot was applied to detect the expression of ZIC1 protein in stably transfected KLE cell,MTT assay was used to screen out IC50 concentration of oxymatrine.Then the cells were divided into four groups:No load without dosing group(A),no load dosing group(B),transfection without dosing group(C) and transfection dosing group(D).Cell adhesion was tested by MTT.Cell migration was analyzed by wound-healing assay.The apoptosis rates were measured by flow cytometry.Results:Expression of ZIC1 protein was positive in experiment groups,showing a significant down-regulation.Compared with control group,both ZIC1 and oxymatrine can decrease cell adhesion(growth),migration of KLE endometrial cancer cell and induce its apoptosis with statistical significance(P<0.05),and the inhibitory effect on migration increased in a time-dependent manner,and the anti-tumor effect of combination of ZIC1 and oxymatrine was more apparent than the other three groups(P<0.05).Conclusion:The combination of ZIC1 gene and oxymatrine worked effectively in KLE cell proliferation repression and apoptosis induction of endometrial cancer,which may have synergistic effect on tumor-suppressing.
