摘    要:目的:利用从肺腺癌(LUAD)数据集中筛选的差异表达基因(DEGs)、差异表达lncRNA(DElncRNA)及其上游miRNA,构建LUAD的竞争内源性RNA网络(ceRNA),探索槲皮素可能的作用靶点。方法:从基因表达综合数据库(GEO)及肿瘤基因组图谱数据库(TCGA)中检索并筛选LUAD基因及lncRNA表达数据集,对差异表达基因(DEGs)进行富集和功能注释。使用在线数据库预测miRNA及lncRNA,建立ceRNA调控网络,并通过网络药理学分析槲皮素的药物靶点。结果:构建了AURKA与上游hsa-miR-363-3p及AP000553.1、LINC00858、AL354707.1的ceRNA调控网络,槲皮素与5DOS对接良好。结论:AP000553.1、LINC00858、AL354707.1与hsa-miR-363-3p竞争性调控AURKA,进一步影响LUAD预后,槲皮素可作用于AURKA。

Abstract:Objective:The differentially expressed genes(DEGs),differentially expressed lncRNA(DElncRNA)and their upstream miRNA,screened from the lung adenocarcinoma(LUAD)data set were used to construct the competitive endogenous RNA network(ceRNA),of LUAD to explore the possible targets of quercetin.Methods:The LUAD gene and lncRNA expression dataset were searched and screened from GEO and TCGA,and the differentially expressed genes(DEGs)was enriched and functionally annotated.Online database prediction miRNA and long non-coding RNA(lncRNA),were used to establish ceRNA regulatory network,and the drug target of quercetin was determined by network pharmacological analysis to explore the molecular mechanism of quercetin in the treatment of LUAD.Results:The ceRNA regulatory networks of AURKA,upstream hsa-miR-363-3p,AP000553.1,LINC00858 and AL354707.1 were constructed,and quercetin was well docked with 5DOS.Conclusion:AP000553.1,LINC00858,AL354707.1 and hsa-miR-363-3p competitively regulate AURKA,and further affect the prognosis of LUAD.Quercetin can act on AURKA.
