子宫内膜样癌及其前驱病变YAP1、PTEN和eIF4E的表达及意义

摘    要:目的:观察子宫内膜样癌及其前驱病变Yes 相关蛋白1 ( YAP1 ) 、张力蛋白同源基因(PTEN)和真核细胞翻译起始因子4E(eIF4E)的表达变化,探讨三者在子宫内膜样癌发生发展中的作用。方法:采用免疫组化SP法分别检测增殖期子宫内膜、单纯性增生 、复杂性增生、子宫内膜上皮内瘤变(EIN)及子宫内膜样癌中的 YAP1、PTEN和eIF4E的表达水平。 结果: YAP1和eIF4E在子宫内膜样癌中的阳性表达显著高于单纯性增生和复杂性增生子宫内膜( P<0.05),PTEN在子宫内膜样癌中的缺失率显著高于单纯性增生和复杂性增生子宫内膜( P <0.05)。YAP1与PTEN阳性表达呈负相关关系(r=-0.420, P<0.05), YAP1与eIF4E阳性表达呈正相关关系(r= 0.818,P<0.05)。结论:从单纯性增生、复杂性增生、 EIN到子宫内膜样癌YAP1和eIF4E的阳性表达率和PTEN的缺失率逐渐增高,三者可能协同参与了子宫内膜样癌的发生发展。

Abstract:Objective: To examine the expression of YAP1,PTEN and eIF4E in endometrioid carcinoma and precursor lesions, and to explore their role in the occurrence and development of endometrioid carcinoma.Methods: Immunohistochemistry SP method was performed to detect the expressions of YAP1, PTEN and eIF4E in proliferative endometrium, simple hyperplasia, complex hyperplasia, endometrial intraepithelial neoplasia (EIN) and endometrioid carcinoma. Results: The expression of YAP1 and eIF4E in endometrioid carcinoma was significantly higher than that in simple hyperplasia and complex hyperplasia endometrium (P <0.05). The loss rate of PTEN in endometrioid carcinoma was significantly higher than that in simple hyperplasia and complex hyperplasia of endometrium (P <0.05). There was a negative correlation between the positive expression of YAP1 and PTEN (r = -0.420, P <0.05), and a positive correlation was found between YAP1 and eIF4E (r = 0.818, P <0.05). Conclusion: The positive rate of YAP1 and eIF4E and the loss rate of PTEN gradually increase all the way from simple hyperplasia, complex hyperplasia, EIN to endometrioid carcinoma, all of which may be involved in the development of endometrioid carcinoma.
