钙蛋白酶-1在胶质瘤组织中的表达及其对U87细胞侵袭和迁移的影响

摘    要:目的 探讨钙蛋白酶-1在人脑胶质瘤组织中的表达及其对U87胶质瘤细胞侵袭和迁移的影响。方法 收集2015年1月至2018年3月手术切除的胶质瘤组织和瘤旁正常脑组织各57例。体外培养U87细胞,将阴性对照小干扰RNA(NC组)和钙蛋白酶-1小干扰RNA(钙蛋白酶-1组)转染至人胶质瘤细胞株U87,以未转染的U87细胞为对照组。采用RT-PCR和免疫印迹法检测mRNA和蛋白表达水平,Transwell实验检测U87细胞侵袭能力,划痕实验评估U87细胞迁移能力。结果 胶质瘤组织钙蛋白酶-1 mRNA和蛋白表达水平均显著高于瘤旁正常脑组织(P<0.05)。钙蛋白酶-1组U87细胞钙蛋白酶-1 mRNA和蛋白表达水平、细胞侵袭数目、细胞迁移率以及转化生长因子-β、基质金属蛋白酶(MMP)-2和MMP-9 mRNA表达水平均明显低于对照组和NC组(P<0.05),而对照组和NC组均无统计学差异(P>0.05)。结论 钙蛋白酶-1在人脑胶质瘤中表达升高,而抑制钙蛋白酶-1表达可能通过影响上皮间质转化,抑制胶质瘤细胞侵袭和迁移。

Abstract:Objective To investigate the changes of Calpain-1 expression in human glioma tissues and the effect of inhibition of Calpain-1 expression on the invasion and migration of cultured U87 cells. Methods The mRNA and protein expressions of Calpain-1 in glioma tissues and normal brain tissues adjacent to the glioma were detected in 57 patients with glioma who underwent microsurgery from January 2015 to March 2018 using RT-PCR and Western blotting, respectively. U87 cells were cultured in vitro, and negative control small interfering RNA (NC group) and Calpain-1 small interfering RNA (Calpain-1 group) were transfected into the U87 cells, with untransfected U87 cells as the control group. The mRNA and protein expressions of Calpain-1 in U87 cells were detected using RT-PCR and Western blotting, respectively. The Transwell test was used to detect the invasion ability of U87 cells, and the scratch test was used to evaluate the migration ability of U87 cells. Results The expression levels of Calpain-1 mRNA and protein in glioma tissues were significantly higher than thos in normal brain tissue adjacent to the glioma (P<0.05). The expression levels of Calpain-1 mRNA and protein, number of cell invasion, cell migration rate and the expression of transforming growth factor-β, matrix metalloproteinase (MMP) -2 and MMP-9 mRNA in U87 cells of Calpain-1 group were significantly lower than those in the control and NC groups (P<0.05),but there was no statistical difference between the control group and the NC group (P>0.05). Conclusion Calpain-1 expression is increased in human gliomas, and inhibiting Calpain-1 expression may inhibit glioma cell invasion and migration by affecting epithelial-mesenchymal transition.
