含粉防己碱生物材料对体外培养软骨细胞的影响

摘    要:[目的]研究含粉防己碱生物材料对体外培养关节软骨细胞的影响。[方法]制备空白对照聚乳酸膜和含粉防己碱的聚乳酸膜。在两种材料表面培养软骨细胞,测定载粉防己碱聚乳酸膜和空白对照聚乳酸膜表面培养软骨细胞的增殖和细胞活性。利用全细胞酶联免疫吸附试验(CellELISA)测试溴代尿苷在DNA合成过程中的掺入量和软骨细胞的Ⅱ型胶原分泌量,测定不同材料表面培养软骨细胞的差别。通过MTT试验测试细胞活性,测定糖胺聚糖分泌量评价软骨细胞功能表达。[结果]细胞形态、活性和组织化学分析结果都表明含有粉防己碱的聚乳酸材料能够促进软骨细胞的增殖和细胞功能表达。[结论]含有粉防己碱的聚乳酸生物材料适用于软骨组织的修复重建和软骨组织工程。

Abstract:[Objective] To investigate the influence of Radix Stephaniae Tetradrae (tetrandrine)-contained biomaterials on chondrocytes in vitro. [Methods] PLLA films with or without tetrandrine were prepared. The proliferation and viability of bovine articular chondrocyte seeded on both films were examined. With whole cell enzyme-linked immunosorbent assay (Cell ELISA) the BrdU incorporation during DNA synthesis and the secretion of collagen type II of the chondrocytes on each PLLA films were detected. Cellular viability was estimated by the MTT assay and the cellular function was assessed by measuring glycosaminoglycan (GAG) secreted by chondrocytes. [Results] The cellular morphology, viability and histochemical results revealed that PLLA materials containing tetrandrine could improve the chondrocyte proliferation and its functions. [Conclusion] The PLLA materials containing tetrandrine are suited to the cartilage reconstruction and the cartilage tissue engineering
