电针对围绝经期失眠大鼠睡眠觉醒脑电图及血清性激素水平的影响

摘    要:目的:观察电针对围绝经期失眠大鼠睡眠觉醒脑电图及血清性激素水平的影响。方法:取40只SD雌性大鼠,按照随机数字表法分为对照组、模型组、电针组和假电针组,每组10只。采用双侧卵巢切除术复制模型,四组大鼠均行脑电图记录电极埋置术,术后恢复10 d,按分组接受不同的干预处理。通过阴道涂片观察大鼠动情周期,利用睡眠生物解析系统客观监测大鼠24 h睡眠觉醒脑电图,ELISA法测定血清促卵泡素(FSH)、黄体生成素(LH)、雌二醇(E2)水平。结果:与对照组比较,模型组、电针组和假电针组大鼠动情周期紊乱率显著增高(P<0.05,P<0.01),模型组和假电针组大鼠血清FSH、LH水平升高(P<0.01)、E2水平降低(P<0.01),电针组大鼠血清FSH升高(P<0.05)、E2水平降低(P<0.01); 模型组和假电针组大鼠24 h睡眠觉醒昼夜节律减弱,睡眠觉醒累积量在白天和全天觉醒增多、非快速动眼睡眠(NREMs)减少(均P<0.01),NREMs在白天和夜间睡眠片段化明显,短时程次数增多(P<0.05)、长时程次数减少(P<0.05,P<0.01),白天慢波睡眠到觉醒(S-W)之间的转换增多(P<0.05,P<0.01); 与模型组比较,电针组大鼠动情周期紊乱率显著降低(P<0.05),大鼠血清FSH、LH水平降低、E2水平升高(均P<0.05),电针组大鼠24 h睡眠觉醒昼夜节律趋势接近对照组,在白天和全天觉醒减少、NREMs增多(P<0.05,P<0.01),白天和夜间短时程睡眠减少、长时程睡眠增多(P<0.05,P<0.01),白天 S-W之间的转换减少(P<0.05),睡眠维持稳定。结论:电针可改善围绝经期失眠大鼠的睡眠觉醒,通过减少觉醒时间,增加睡眠时间,并改善其睡眠结构,其机制可能与调节血清性激素水平有关。

Abstract:Objective:To observe the effect of electroacupuncture(EA)on sleep arousal electroencephalogram(EEG)and serum sex hormone levels in perimenopausal insomnia rats.Methods:A total of 40 SD female rats were divided into four groups:control,model,EA and sham EA group according to random number table method,with 10 rats in each group.Bilateral oophorectomy was used to replicate the model.EEG recording electrode implantation was performed on all the rats in four groups.Postoperative recovery was 10 days,different intervention treatments were performed in different groups.The estrus cycle of rats was observed by vaginal smear,24-hour sleep and wakefulness EEG was monitored objectively by sleep bioanalytic system,and serum levels of FSH,LH and E2were determined by ELISA.Results:Compared with the control group,the disturbance rate of estrous cycle was significantly increased in model group,EA group and sham EA group(P<0.05,P<0.01),serum FSH and LH levels in model and sham EA group were increased(P<0.01)and E2levels were significantly decreased(P<0.01),while serum FSH levels in EA group were increased(P<0.05)and E2levels were significantly decreased(P<0.01).In the model and sham EA group,the circadian rhythm of 24-hour sleep awakening was weakened,the cumulative amount of sleep arousal increased and NREMs decreased in the daytime and the whole day(P<0.01).NREMs showed significantly sleep fragmentation in daytime and nighttime,increased frequency of short duration(P<0.05),decreased frequency of long duration(P<0.05,P<0.01),and increased conversion from slow wave sleep to wake(S-W)in daytime(P<0.05,P<0.01).Compared with the model group,the estrous cycle disturbance rate of rats in the EA group was significantly decreased(P<0.05),the levels of serum FSH and LH were decreased,and the level of E2 was increased(P<0.05).The circadian rhythm trend of 24 h sleep awakening in EA group was similar to that in the control group,awakening decreased and NREMs increased in day and whole day(P<0.05,P<0.01),short duration sleep decreased and long duration sleep increased in daytime and nighttime(P<0.05,P<0.01).During the daytime the conversion between S-W decreased(P<0.05),and the sleep remained stable.Conclusion:Electroacupuncture can improve the sleep arousal of perimenopausal rats with insomnia by reducing the wake time,increasing the sleep time,and improving the sleep structure.The mechanism may be related to the serum sex hormone levels.
