兔骨髓间充质干细胞向成骨方向的诱导分化及体内应用

摘要: 目的:研究经矿化诱导的兔骨髓间充质干细胞(MSC)在裸鼠体内的成骨潜能。方法:分离并培养兔MSC,矿化诱导,观察MSC的形态学改变及碱性磷酸酶(ALP)活性。将诱导后的MSC-去抗原牛松质骨复合体植入裸鼠背部皮下,3 d后取材,行组织学观察。结果:经矿化诱导后,MSC的形态由长梭形向多角形和三角形转化,ALP组织化学染色阳性,活性明显增高。MSC-去抗原牛松质骨复合体植入裸鼠后3 d,组织学见细胞均匀贴附于骨小梁表面,伸展良好,未见未贴壁细胞及外源性纤维组织、血管长入。结论:经矿化诱导的MSC向成骨细胞方向分化和增殖,经观察MSC-去抗原牛松质骨复合体在裸鼠体内可成活并增殖。

Abstract: Objective\ To study the osteogenic property of the induced rabbit bone messenchymal stem cells(MSC) in nude rats. Methods MSC were obtained by using gradient centrifuge method. The cells had been cultured in mineralized medium for 1 week, then the ALP activity was observed. The induced MSC/antigen-extracted bovine cancellous bone were implanted in the nude mice subcutaneously. At 3rd day after implantation, the implants underwent the histological analysis. Results ALP activity was higher and the cells displayed fibroblast-like morphology, revealed positive staining with alkaline phosphatase method in the induced medium. At 3rd day after implantation, histological analysis revealed that the induced cells were adhered at the surface of the bone trabeculase and growed well. Exogenous fibro-tissue, blood vessel and free induced cells were invisible. Conclusion Rabbit MSC could differentiate into osteoblast and proliferate induced by the mineral medium. The induced MSC/antigen-extracted bovine cancellous bone could survive in the nude rat.
