Recent progress on Wnt pathway and osteosarcoma

摘要: 骨肉瘤是青少年及儿童最常见的原发性恶性骨肿瘤, 40%的患者预后不佳。有研究发现, Wnt信号传导通路在骨细胞的增殖、分化和凋亡中起重要作用。这条通路在骨量的调节过程中有着关键性的意义。其通过一系列调控蛋白进行功能调节。通路调控异常引发其组成成分的突变, 导致了一系列骨疾病, 特别是骨肉瘤的发生。Wnt信号通路相关靶向治疗药物的应用, 可以使骨肉瘤的治疗得到更好的发展, 并且这类药物已逐渐受到人们的重视。

Abstract: Osteosarcoma is the most prevalent primary malignant bone tumor in children and teenagers with a poor survival in 40% of the victims.The Wnt/β-catenin signaling pathway has been shown to play a major role in bone cell differentiation, proliferation and apoptosis.It is a critical component of bone mass regulation and required for bone to respond to mechanical loading.The pathway is tightly regulated by a number of modulator proteins.Mutations in pathway components that result in aberrant regulation are involved in a number of bone diseases, especially Osteosarcoma.The use of Wnt pathway associated markers appear promising for osteosarcoma.Drugs targeting the Wnt pathway will gain even more attention.
